My Camera won't upload photos

I have a Nikon, D3000, and whenever I put my USB into the laptop, turn the computer on so it will do it's thing and whenever is says "Locating Photos and Videos" then i get a message saying that "No new photos/videos could be located/found" and I JUST took some pictures.
Whenever I tried this on our Desktop, it don't even load anything or move when i plug it in, it just makes a little "Doo-Dum" noise to let me know I plugged it in, and that's it. Help!

Added (1). Did i mention when I slide the memory card to into the card reader… IT ALSO DOES NOTHING?

My guess is that you are NOT following the instructions in your user manual

But what you will find works better is to remove the memory card from the camera and place it into a card reader. This is what we have been doing for over a decade. Just use your computers file manager to copy all the images from your memory card to a new file folder on your computer named for the subject and date. Sample: InsuranceShots072012

You now have a fully adjustable camera, so you need to learn how to control every aspect of the digital photographic process, not depending upon the camera or computer do the work for you.

To properly use the USB cable, you generally have to load an app from Nikon.

At some point, you may have to use Windows Explorer to navigate the camera, and/or nix the USB cable and just remove the SD card from the camera and plug it into your computer. This is the method most often used.

Your desktop has an incompatible driver.

These are computer issues, not camera problems, so troubleshoot it from that perspective.

May be it is fault with the memory card or you need to check it with the technician who understands it well.

Do not use the computer's built-in card reader. Buy a cheap one (with emphasis on the cheap part for better compatibility). Use that to connect the camera's memory card directly to an empty USB slot in your computer. This method eliminates all camera to computer connection problems. After the Doo-Dum, use Windows Explorer or (Mac) Finder to do the actual Copy for absolute total control of what goes where exactly.
